三七皂苷Rb1改善2型糖尿病大鼠胰岛素抵抗的作用研究 被引量:4

摘要 目的:构建2型糖尿病大鼠模型,探讨三七皂苷Rb1在2型糖尿病大鼠胰岛素抵抗中的作用及其机制。方法:高脂、高胆固醇灌胃+腹腔注射链脲佐菌素构建2型糖尿病大鼠模型,同时给予三七皂苷Rb1治疗,检测血清糖化血清蛋白(FMN)、甘油三酯(TG)、总胆固醇(TCHOL)、超敏C反应蛋白(CRP)、血糖(FGLU)、胰岛素(INS)、C肽(C-P)的含量和丙二醛(MDA)、谷胱甘肽(GSH)、谷胱甘肽还原酶(GR)的含量。结果:模型组FMN、TG、TCHOL、CRP、C-P、INS和IRI明显高于对照组(P<0.05);模型组的T-SOD、GSH和GR的含量明显低于对照组(P<0.05);而MDA明显高于对照组(P<0.05)。经三七皂苷Rb1治疗后,明显改善模型大鼠高脂血症和胰岛素抵抗,提高模型大鼠内源性巯醇抗氧化物酶的含量和减少肝组织的脂质过氧化。结论:三七皂苷Rb1能通过调控肝组织内源性巯醇抗氧化物(酶)的含量,改善2型糖尿病中的IR。 Objective: The objective of the present study was to test whether Notoginsenoside Rbl contribute to the pathogenesis of insulin resistance in type 2 diabetes. Methods: Male Sprague - Dawley rats (210 ± 10) g were divided into three groups and fed with commercially available normal diet or in - house prepared high - fat diet ( HFD ) ( composedby 250ml pork fat and 5g Cholesterol) for a period of 3 months. After 3 months of dietary manipulation, a subset of the rats from model groups was injected intraperitoneally with low dose of streptozotoein (STZ, 20 mg/kg) two times andfed to normal diet for l months again. Results: The model rats showed significant increasing FGLU, CRP, INS, TG, TCHOL, IR and GLU, INS , C -P in OGTF -2 hour as compared to ND rat. At the same time, Chronic systemic exposure of HFD + STZ to rats induced insulin resistance and increased in liver oxidative stress, including an increase in malondialdehyde (MDA), Glutathionereduetase (GR) (P 〈0. 05), while a decrease in Nitric glutathione (GSH) (P 〈 0.05 ) . The significant deviation was not detected in total total superoxide dismutase ( T - SOD) . After notoginsenoside Rbl treat- ment, significant improvement in rat model of hyperlipidemia and insulin resistance, improve the content of endogenous Thiol antioxi- dant enzymes in the rat model and reduction of liver tissue lipid peroxidation. Conclusion: Notoginsenoside Rbl can induce insulin re- sistance by manipulating thiol antioxidants, GR and GSH of the liver in rats.
