摘要
目的探讨高血压患者血浆同型半胱氨酸(Hcy)水平及亚甲基四氢叶酸还原酶(MTHFR)A1298C基因多态性对血浆Hcy水平的影响。方法选择原发性高血压患者145例,采用荧光偏振免疫法测定血浆Hcy水平,依据Hcy水平将研究对象分为H型高血压组及非H型高血压组。并应用聚合酶链反应-限制性内切酶片段长度多态性技术检测MTHFR基因多态性。结果 H型高血压组及非H型高血压组血浆平均Hcy水平分别为(16.74±6.14)μmol/L,(8.50±1.02)μmol/L,两者差异有统计学意义(P<0.05)。H型高血压组及非H型高血压组MTHFRA1298C多态位点AA、AC及CC基因型频率分布及C等位基因频率差异均无统计学意义(P>0.05)。AA、AC和CC 3种基因型间的平均血浆Hcy水平比较差异无统计学意义(P<0.05)。结论 H型高血压患者血浆Hcy水平较非H型高血压显著升高,MTHFR A1298C多态位点对血浆Hcy水平无明显影响。
Objective To investigate homocysteine levels in patients with hypertension and the relationship between plasma homocysteine(Hcy) and methylenetetrahydrofolate reductase(MTHFR)gene polymorphisms.Methods 145 hypertension patients were selected.Plasma Hcy was measured by fluorescence polarization immunoassay(FPIA).Acording to the homocysteine levels,the patients were divided into H-type hypertension group and common hypertension group.Polymorphisms of the MTHFR gene were measured by Polymerase chain reaction and restriction fragment length polymorphism(PCR-RFLP).Results Plasma homocysteine levels was significantly higher in H-type hypertension group than in common hypertension group((16.74±6.14)μmol/L VS(8.50±1.02)μmol/L,P0.05).The genotype distribution frequencies of MTHFR A1298C between the two groups were not significant differences(P0.05).Homocysteine levels were not significant different among the three genotype AA,AC and CC(P0.05).Conclusion Hmocysteine level was significantly higher in H-type hypertension group than in common hypertension group.MTHFR A1298C mutation has no significant effect on plasma homocysteine levels.
