摘要
目的通过奥氮平处理心肌细胞来探讨奥氮平诱导心血管疾病的机制。方法用不同浓度(10,20和40uM)及不同作用时间(1,3,12和24 h)的奥氮平处理大鼠心肌细胞H9C2,检测巨噬细胞迁移抑制因子(Macrophage migration inhibitory factor,MIF)在mRNA及蛋白水平的表达。结果用不同浓度、不同时间的奥氮平处理H9c2细胞时,随着处理浓度的增加及时间的延长,MIF mRNA及MIF蛋白随着浓度的递增及时间的延长而增高(P<0.05)。结论奥氮平处理心肌细胞后,MIF的mRNA及蛋白均发生显著改变,这种改变存在浓度与时间的依赖关系,MIF可能参与奥氮平诱导的心血管系统损伤。
Objective To examine the effects of olanzapine on macrophage migration inhibitory factor(MIF)expression in cardiomyocytes.Method Rat H9C2 cardiomyocytes were cultured and treated by olanzapine with different concentrations from 10 to 40 uM for different time course from 1 to 24 h.The mRNA and protein expression levels of MIF were measured at each concentration and time point.Result Both MIF mRNA and protein levels increased following olanzapine treatment in H9C2 cardiomyocytes(P<0.05)in time and dose dependent manners.Conclusion Olanzapine can increase MIF expression in cardiomyocytes,suggesting that olanzapine treatment may regulate MIF signaling and potentially affect the development of cardiovascular diseases.
