惠楼山药内生细菌的分离和特性研究 被引量:3

Study on Isolation and Characteristic of Endophytic Bacteria in Huilou Chinese Yam

摘要 对惠楼山药块茎不同部位的内生菌进行分离纯化,采用革兰氏染色、芽孢染色法,结合菌落特征和培养特征将其内生细菌初步分为两大类,即第Ⅰ类型、第Ⅱ类型。采用浓度梯度稀释涂布法分别进行微生物计数。试验结果表明,内生菌的总体数量分布为块茎下部>块茎上部>块茎中部,说明内生细菌在山药块茎中的分布因部位不同存在差异。基于染色特征、培养特征和生理生化培养特性,第一类型符合芽孢杆菌属(Bacillus)的特点,第Ⅱ类型符合叶瘤杆菌属(Phyllobacterius)的特点。研究山药内生菌的分布和类型,筛选出代谢产生山药多糖的内生细菌,在医疗、保健方面将有重要的科学和应用价值。 Endophytic bacterias of tuber in different parts of Huilou Chinese Yam were isolated and purified. They were classified into two types, ie, type I and type 11 based on Gram dye and Spore dye methods combined clony characteristic and cultive characteristic. Microbes were counted by concentration gradient dilution spread method. Results showed that the distribution of endophytic bacteria total number in tuber was under-part 〉tuber up-part〉tuber mid-part. It illustrated that the difference between endophytic bacteria distribution in tuber was in accordance with different parts. Based on dye features, cultive characteristic and physiological and biochemical characteristics, endophytic bacterial type l accorded with genus Bacillus, and endophytic bacterial type Ⅱ accorded with genus Phyllobacterius. It was of important science and application values in medical treatment and health care to study the distribution and type of endophytic bacteria, and to screen endophytes producing Yam polysaccharides.
