高糖对RF/6A细胞内PI3K-Akt信号通路的影响 被引量:12

The effect of high glucose on PI3K-Akt signal pathway in RF/6A cells

摘要 目的探讨高糖对视网膜血管内皮细胞内血管新生关键信号通路PI3K-Akt信号通路的影响。方法以猴视网膜血管内皮细胞RF/6A细胞为研究对象,分为空白对照组、甘露醇对照组、高糖组和PI3K-Akt信号通路特异性阻断剂LY294002组,分别采用MTs法检测RF/6A细胞增殖、划痕实验检测细胞迁移以及Matrigel法检测细胞管腔形成能力,Western blot法检测PI3K-Akt信号通路中关键蛋白Akt的表达以及磷酸化。结果与空白对照组相比,甘露醇对照组细胞增殖率、迁移率、管腔形成数以及细胞内Akt及p-Akt的表达无统计学差异(P>0.05);高糖组细胞增殖增加15.12%(P<0.05),细胞迁移增加39.24%(P<0.01),管腔形成数增加59.10%(P<0.01),细胞内Akt及p-Akt的表达分别增加27.34%(P<0.05)、25.23%(P<0.05);LY294002组细胞增殖减少17.56%(P<0.05),细胞迁移减少45.71%(P<0.01),管腔形成减少39.08%(P<0.01),Akt及p-Akt的表达量分别减少37.78%(P<0.01)、42.08%(P<0.01);与高糖组相比,LY294002组细胞增殖减少29.06%(P<0.05),细胞迁移减少61.01%(P<0.01),管腔形成减少62.14%(P<0.01);Akt及p-Akt的表达分别减少51.15%(P<0.01)、53.75%(P<0.01)。结论高糖可经上调RF/6A细胞内PI3K-Akt信号通路促进血管新生,PI3K-Akt信号通路阻断剂可拮抗高糖的促RF/6A细胞血管新生的作用。 Objective To study the effect of high glucose on PI3K-Akt signal pathway in rhesus retinal endothelial RF/6A cells.Methods RF/6A cells were treated with PBS,mannitol,high glucose and LY294002 respectively.The MTS,cell migration and lumen formation experiments were used to detect cell proliferation,migration and lumen formation respectively.Western blot was used to measure the expression of both Akt and p-Akt.Results There was no significant difference in cell proliferation,migration,lumen formation and the expression of both Akt and p-Akt between the blank control group and manitolcontrol group(all P0.05).High glucose increased cell proliferation,migration,lumen formation by 15.12%,39.24%,59.10%respectively(P0.05 or P0.01),and the expression of Akt and p-Akt enhanced by 27.34% and 25.23%(both P0.05),while LY294002 decreased cell proliferation,migration,lumen formation by 17.56%,45.71%,39.08%respectively(P0.05 or P0.01),and the expression of Akt and p-Akt reduced by 37.78% and 42.08%(both P0.01).Compared to high glucose group,cell proliferation,migration,lumen formation decreased by 29.06%,61.01%,62.14%respectively in LY294002 group(all P0.01)and the expression of Akt and p-Akt reduced by 51.15% and 53.75%(both P0.01).Conclusions High glucose could up-regulate PI3K-Akt signal pathway to promote RF/6A cells angiogenesis.Blockers of the pathway could significantly suppress this effect stimulated by high glucose.
