白藜芦醇对MG132诱导K562细胞凋亡影响的研究 被引量:1

Effect of resveratrol on apoptosis induced by MG132 of K562 cells

摘要 目的:研究白藜芦醇能否增加K562细胞对MG132的敏感性。方法:MG132单独及联合白藜芦醇作用于K562细胞,MTT法检测细胞活力,流式细胞仪检测细胞凋亡,蛋白质印迹法检测PARP剪切水平。结果:>50μmol/L白藜芦醇能够有效地抑制K562细胞的活力,P<0.01;MG132单独处理K562细胞24h,对细胞活力的抑制呈剂量依赖性,P<0.01;而联合用药时,白藜芦醇呈剂量依赖性的对抗了MG132对K562细胞的毒性,并且5μmol/L白藜芦醇有效的降低了K562细胞凋亡率,P<0.05。结论:白藜芦醇具有对抗MG132诱导细胞毒性的潜在作用。 OBJECTIVE: To investigate whether resveratrol could sensitize K562 cells to MG132.METHODS: K562 cells were treated with MG132 alone or in combination with resveratrol.Cell viability was checked by MTT assay,and induction of apoptosis was measured by flow cytometry.Western blot was used to investigate the level of PARP cleavage.RESULTS: Viability of K562 cells was inhibited after treatment with resveratrol's concentration more than 50 μmol/L(P0.01).Viability of K562 cells treated with MG132 demonstrated a dose dependent decrease within 24 h(P0.01).But when combination with MG132,resveratrol significantly prevented the cytotoxicity induced by MG132 in a dose dependent manner.And 5 μmol/L resveratrol strongly reduced apoptosis rate of K562 by MG132(P0.05).CONCLUSIONS: Resveratrol may have the potential to negate the cytotoxic effects induced by MG132.
