摘要
为制备抗鸡传染性喉气管炎病毒(ILTV)糖蛋白gD的单克隆抗体(MAb),本研究通过原核表达gD重组蛋白,纯化后免疫6周龄雌性BALB/c小鼠,经细胞融合筛选获得一株稳定分泌抗ILTV gD蛋白的杂交瘤细胞株,MAb亚型经鉴定为IgG1,轻链为κ链。Western blot结果显示,这株杂交瘤细胞分泌的MAb能够识别ILTV。ILTV gD蛋白的MAb的制备,为ILTV检测方法的建立奠定了基础。
To prepare monoclonal antibody (MAb) against glycoprotein D of infectious laryngotracheitis virus (ILTV), the SP2/0 cells were fused with spleen cells of BALB/c mice immunized with the purified recombinant gD protein, a hybridoma cell line stably secreting antibody was obtained by scanning with indirect ELISA coating with purified ILTV as coating antigen. The MAb was identified to be IgG1 subclass with K light chain. Western blot analysis showed that MAb reacted positively with ILTV. The MAb developed could serve as a tool for further study on ILTV and development of detection method for ILTV infection.
