凉膈散对肠缺血再灌注损伤大鼠肠道的保护作用 被引量:12

The Protective Effect of LIANGGE POWDERS on Intestinal Ischemia/Reperfusion Injury in Rat

摘要 目的:观察凉膈散对大鼠缺血再灌注(I/R)损伤肠黏膜的保护作用。方法:健康Wistar大鼠168只,随机分为正常组、假手术组、模型组、凉膈散治疗组和凉膈散防治组,其中正常组8只,其他4组各40只。正常组不予任何处理,假手术组只分离不夹闭肠系膜上动脉,其余3组通过夹闭肠系膜上动脉建立大鼠小肠I/R模型,根据缺血45min后再灌注3、12、24、48和72h5个时相点又分为5组,每组8只大鼠。测定各组5个时点动物小肠黏膜病理损伤Chiu氏评分、门静脉血浆D-乳酸值、小肠组织匀浆丙二醛(MDA)含量和超氧化物歧化酶(SOD)活力、肠黏膜细胞凋亡指数及Bax表达水平。结果:模型组动物肠黏膜结构破坏明显,Chiu氏评分、血浆D-乳酸值明显升高。小肠匀浆MDA含量升高、SOD活力降低,肠黏膜细胞凋亡指数及Bax表达水平升高。凉膈散干预后,可减轻小肠黏膜病理损伤程度、与模型组比较,除3h外,门静脉血浆D-乳酸值下降,小肠组织匀浆MDA含量降低和SOD活力升高、肠黏膜细胞凋亡指数及Bax表达水平均下降。结论:I/R损伤可破坏肠黏膜屏障功能,凉膈散可通过减少机体氧自由基的生成而保护肠黏膜。 Objective: To observe the protective effect of LIANGGE POWDERS on intestinal ischemia/reperfusion (I/R) injury of rats. Methods: A total of 168 Wistar rats were randomly divided into 5 groups: normal group, sham operation group, model group, LIANGGE POWDERS therapeutics group and LIANGGE POWDERS prevention and treatment group. There were 8 rats in normal group and 40 in other 4 groups. The I/R rat models were established by blocking the superior mesenteric artery in rats of sham operation group, model group, LIANGGE POWDERS therapeutics group and LIANGGE POWDERS pre- vention and treatment group. The model rats were subdivided into 5 groups, 3 h, 12 h, 24 h, 48 h and 72 h reperfusion after 45 min ischemia (n=8 for each). The values of Chiu' s score, D-lactic acid content of janitrix blood plasma, malondialdehyde (MDA) content and superoxide dismutase (SOD) activity of intestinal homogenate, apoptotic index and the expression of the apoptotic protein Bax in the small intestine were measured in 5 subgroups. Results: After I/R, the structure of the intestinal mucosa was significantly injured. The values of Chiu' s score and D-lactic acid content of blood increased significantly. The intestinal MDA content increased and SOD activity decreased in rats of model group. The apoptotic index and the expression of Bax were obviously higher in intestinal mucosal cells of model group than those in other groups. After the intervention of LI- ANGGE POWDERS, the Chiu' s score and D-lactic acid content were decreased than those in model group. The value of D-lactic acid and MDA content decreased and SOD activity increased in subgroups except for that of 3 h group. The apoptotic index and the expression of Bax dropped as well. Conclusion: The I/R injury can destroy the function of intestinal barrier, and LIANGGE POWDERS can protect the intestinal mucosa through cleaning the oxygen free radical.
