摘要
用分泌抗人绒毛膜促性腺激素(hCG)单克隆抗体的6-巯基鸟嘌呤抗性杂交瘤突变株BAH_1-TG细胞与用辣根过氧化物酶(HRP)免疫的小鼠脾细胞进行二次杂交。二次杂交瘤细胞的培养上清用双抗原夹心放射免疫测定法和双抗原夹心酶联免疫测定法检测抗hCG-抗HRP双特异抗体。经多次克隆和筛选,获得两株分泌抗hCG-抗HRP双特异单克隆抗体的二次杂交瘤株E_4和E_7。
The BAH1-TG mutant secreting anti-hCG antibody, which is screened by 6-TG and sensitive to HAT, was hybridized with spleen cells of BALB/c mice immunized with HRP. The superna-tants of hybrid hybridomas were secreened by double-antigen sandwich RIA and ELISA. By using microtechnique and limiting dilution cloning, two lines of hybrid hybridomas secreting anti-hCG-anti-HRP bispecific monoclonal antibodies were obtained, which were named E4 and E7 respeclively.
