摘要
目的分析西宁市2011年麻疹病毒流行株的基因特征。方法用Vero/slam细胞从发热伴出疹病例的咽拭子标本中分离出麻疹病毒,采用逆转录-聚合酶链反应-限制性片段长度多态性分析(Reverse Transcription-Polymerase Chain Reaction-Restriction Fragment Length Polymorphism,RT-PCR-RFLP)方法,检测毒株。结果应用H1基因型麻疹野病毒RT-PCR-RFLP方法,对麻疹毒株进行鉴定,其RT-PCR阳性产物经Sal I酶切后被切成2个片段,分别为408个和159个核苷酸,显示该病例由H1基因型麻疹病毒引起。结论 2011年分离到的5株麻疹毒株均为H1基因型麻疹病毒,与中国其它地区的流行情况一致,无新型及传入性的其它亚型出现。
Objective To investigate the genetic characteristics of measles virus in Xining City in 2011.Method Adopting Vero/SLAM cell to isolate measles virus from throat swab specimens of fever with eruption,and detected by Reverse Transcription-Polymerase Chain Reaction-Restriction Fragment Length Polymorphism method(RT-PCR-RFLP).Results Measles virus strain was identified by H1 genotype of wild measles virus of RT-PCR-RFLP method.There were 2 nucleotide fragments when RT-PCR positive products enzyme-cut by Sal I,including 408 and 159 nucleotide respectively,which indicate the cases caused by H1 genotype of measles virus.Conclusions In 2011,5 measles virus strains were isolated,and all were H1 genotype of measles virus,which accord with the epidemic status of other areas in China,no new type and other foreign subtype appeared in 2011.
