摘要
利用头孢菌素和反离子HPO_4^(2-)的相互作用,在匍聚糖凝胶Sephadex G-10色谱柱中,对12种不同结构的头孢菌素中的高分子杂质进行了分离。 离子对试剂可以明显改变头孢菌素在葡聚糖凝胶Sephadex G-10中的色谱行为,使其Kav值增大,改变幅度和头孢菌素的侧链结构有关。依据实验结果,对反离子HPO_4^(2-)和头孢菌素作用,改变头孢菌素在Sephadex凝胶中色谱行为的机制进行了讨论。
We have found that the interaction between HPO_4~2 and cephalosporins is strongly changing the chromatographic behaviour on Sephadex G-10, while using this phenomena, the high molecular weight impurities of cephalosporins were isolated.Ion-pair reagent could increase the Kav value of cephalosporins, and which in turn had close relationship with the substitution group of the antibiotic.Based on the experimental results, the mechanism of ion-pair reagent which influenced the cephalosporin chromatographic behaviour were also discussed.
