摘要
建立了催化动力学荧光光度法测定辣根过氧化物酶(HRP)及人甲胎蛋白(AFP)的新方法。在含TritorX-100的磷酸盐缓冲溶液中,辣根过氧化氢氧化邻苯二胺的产物2,3-二氨基吩嗪的荧光强度有明显的增强。在一定的实验条件下,该产物的荧光强度与HRP的量成正比,方法对HRP的线性范围为10~200pg,线性相关系数为0.9956,最低检出下限可达8pg,其相对标准偏差为3.2%(n=10)。
A novel fluorimetry for the determination of horseradish peroxidase(HRP) and α-fetoprotein (AFP) is developed. In the phosphate buffer containing TritonX-100, the fluorescence signal of 2, 3-dia minophinazine which is an oxidized product of o-phenylenediamine catalyzed by HRP, was enhanced.Under certain experimental condition, the fluorescence intensity of the product is proportional to the amount of HRP, the linear range for HRP is 10 ~ 200 pg/tube with a correlation coefficient of 0. 9956.The detection limit of HRP is 8 .0 pg/tube and the RSD for 20 pg/tube is 3.2% (n = 10). The α-feto-protein in human serum was determined by classical enzyme-linked immunoassay detected by this method with a satisfactory result.
