蛋白质分离与纯化过程中高效离子交换色谱柱的再生 被引量:2

摘要 高效离子交换色谱法分离和纯化蛋白质时,蛋白质在色谱柱上的不可逆吸附使柱效降低,柱压升高。本文使用2%的胰蛋白酶溶液在37℃下处理TSDgal CM-5PW和TSKgelDEAE-5PW(75×7.5mmI.D.)离子交换色谱柱24h后,柱压显著降低,柱效提高4~13倍,交换容量也得到恢复。离子交换色谱术的恢复程度和交换容量与处理液的用量有关。 Irreversible adsorption of proteins on high-performance ion-exchange packings can result in column deterioration and damage. TSKgel CM-5PW column and a DEAN-5PW (75 × 7. 5 mm I. D. ) column fouled by irrevesible adsorption of proteins were treated with 2% trypsin solution under 37℃ for 24 hours, the back pressure was significantly declined, the number of theoretical Plate was increased by 4 to 13 times and ion-exchange capacity was restored. The extent of column regeneration was related with the volume and concentration of the trypsin solution.
