摘要
【目的】建立糖尿病兔模型,通过超声心动图观察糖尿病兔心脏结构、血流动力学及心功能的变化。【方法】通过静脉注射致病剂量链脲左菌素(sTz)配合高脂饮食,诱发兔高血糖,制备2型糖尿病兔模型。应用超声心动图分别观测正常组(NC组,20只)与糖尿病组(DM组,30只)的心脏结构变化以及心功能改变。【结果1DM组给药4周后左房前后径(LAd)、左室舒张末期内径(LVDD)、舒张期室间隔厚度(IVSd)、左室后壁厚度(LVPwd)均明显高于NC组(P〈O.05)。DM组给药8周后左室射血分数(EF)及左室短轴缩短率(FS)较NC组降低(P〈0.05)。DM组给药2周后心肌活动指数(MPI)又称Tei指数,较NC组进行性增大。【结论】通过应超声心动图能够早期、定量、准确的观测2型糖尿病兔心功能异常改变,为临床早期检测糖尿病心脏病变提供了安全、可靠、无创的新途径。
[Objective]To establish the rabbit model with type 2 diabetes mellitus(T2DM), and observe the changes of cardiac structure, hemodynamics and cardiac function by using echocardiography. [Methods] Rabbit model with T2DM was prepared by inducing hyperglycemia through intravenous injection of streptozo tocin(STZ) and highfat diet. Echocardiography was used to observe the changes of cardiac structure and func tion in the normal group(NC group, n =20) and diabetic group (DM group, n =30). [Results]Left atrium di ameter(LAd), left ventricular diastolic inner diameter (LVDd), interventricular septum diameter (IVSd) and left ventricular posterior wall thickness(LVPWd) in DM group were obviously higher than those in NC group ( P d0.05). Compared with NC group, left ventricuiar ejection fraction(LVEF) and left ventricular short ax is shorten rate(FS) in DM group after 8 weeks of medication were decreased( P d0.05). Compared with NC group, myocardial performance index(MPI or Tei index) in DM group after 2 weeks of medication were pro gressively increased. [Conclusion] Echocardiography can early and accurately observe the normal changes of cardiac function in the rabbits with T2DM in order to provide a safe, reliable, noninvasive and new way for the early detection of diabetic cardiopathy.
