摘要
从云雾龙胆中分离到内生菌LD-b1,通过形态观察、生理生化实验和16SrDNA系统进化分析,鉴定其为短小芽孢杆菌(Bacillus pumilus)。该菌株的发酵上清液对苹果腐烂病菌的生长有显著的拮抗作用,拮抗活性物质对80℃以下的温度有一定耐受能力;在pH5~10之间均有一定活性,对低pH更敏感;50μg/mL的蛋白酶K在37℃条件下处理1h可以使其抑菌活性完全丧失。抑菌曲线测定结果表明,摇瓶发酵48h时内生菌LD-b1的抑菌活性最高,在苹果果实上对腐烂病斑也具有明显的抑制作用。
Endophytic strain LD-b] was isolated from Gentiana nubigena. According to the characteristics of morphology,physiology and biochemistry tests and the comparison of 16S rDNA sequence, strain LD-b1 was identified as Bacillus pumilus.The fermentation broth of LD-b1 showed antagonism activities against Valsa mali.The antibiotic active substance was stable to the temperature lower than 80%,and the pH range from 5 to 10, but more susceptible to lower pH.The antifungla activity lost under the treatment with 50μg/mL proteinase K in the conditions of 37℃ longer than 1h.The results of shaking flask fermentation curves showed antibiotic activities reached the maximum at 48h.The lesion diameter was smaller obviously when apple fruits were treated with LD-bi.
