摘要
建立一种反相高效液相色谱法测定3,5-二甲氧基苯甲醛取代物ND的含量及其有关物质。采用Agilent Zorbax C18(250mm×4.6mm,5μm)色谱柱,流动相为甲醇-0.002%醋酸(40︰60 v/v),流速1.0mL/min,检测波长210nm,柱温35℃,进样量20μL,外标法测定。在该色谱条件下,3,5-二甲氧基苯甲醛取代物ND及其有关物质能够得到较好的分离,分离度大于1.5,理论塔板数按3,5-二甲氧基苯甲醛取代物ND计大于5000;3,5-二甲氧基苯甲醛取代物ND浓度在0.02~0.14mg/mL范围内与峰面积呈良好的线性关系(r=0.9991);检测限和定量限分别为2ng和4ng。该高效液相色谱法准确、灵敏、可靠,专属性好,可用于3,5-二甲氧基苯甲醛取代物ND的含量及其有关物质的测定。
To establish an RP-HPLC method for the determination of the content of derivative of 3,5-dimethoxy benzaldehyde ND and related substances.The analysis was carried on an Agilent Zorbax C18(250mm×4.6mm,5μm) column;The mobile phase was composed of methanol and 0.002% acetic acid(40︰60 v/v)at the flow rate of 1.0 mL/min;The detection wavelength was 210 nm,and the column temperature was 35℃;Loaded volume was 20μL.Using this method,derivative of 3,5-dimethoxy benzaldehyde ND and its related substances were well separated and the degree of separability was more than 1.5,the number of theoretical plates with derivative of 3,5-dimethoxy benzaldehyde ND was more than 5000;The linear cure was obtained in the range of 0.02~0.14mg/mL(r=0.9991);The minimal detectable level and the minimal quantifiable level were 2ng and 4ng.The method proposed for determination of the content and its related substances of derivative of 3,5-dimethoxy benzaldehyde ND is specific,reliable and accurate,and can be used for the determination of the content of derivative of 3,5-dimethoxy benzaldehyde ND and its related substances.
