氧化苦参碱体外抑菌活性的研究 被引量:14

Study on the bacteriostasis effect of oxymatrine in vitro

摘要 目的评估氧化苦参碱对条件性致病菌——大肠埃希菌的抑菌活性。方法采用滤纸片法和96孔板法对氧化苦参碱的抑菌活性进行检测,根据不同浓度氧化苦参碱对大肠埃希菌的抑菌率,利用origin 75拟合标准曲线计算氧化苦参碱对大肠埃希菌IC50。结果滤纸片法检测结果发现,256、128、64、32 mg/mL的氧化苦参碱对大肠埃希菌的抑菌圈大小分别为7.75、7.25、6.50和6.00 mm,而阳性对照5 000 IU/mL庆大霉素的抑菌圈大小为27.00 mm。96孔板法检测结果发现,氧化苦参碱对大肠埃希菌的抑制率均随用药剂量的增加抑制率增大,呈"S"型曲线。用药后6、12、24、48和72 h时氧化苦参碱的IC50值分别为27.27、26.81、24.75、20.29和17.01 mg/mL。结论氧化苦参碱对大肠埃希菌生长有抑制作用,其抑菌作用随氧化苦参碱用药作用时间的延长而增强,但抑菌活性较低。 Objective To evaluate the bacteriostasis effect of oxymatrine against Escherichia coli by using bacterial sensitivity test in vitro. Method The Microplate Reader and filter paper disc were used in this study. The size of inhibition zone was measured 24 hours after different dose of oxymatrine used in the experiment, while gentamycin was used as a positive control and natural saline as a negative control. The curves were plotted and the dose that inhibited 50% Escherichia coli growth ( IC50 ) was determined by using origin 75 software. Result The inhibition zones of fiher paper were 7.75, 7.25, 6.50, 6.00 and 27.00 mm after 256, 128, 64, 32 mg/mL oxymatrine and 5 000 IU/mL gentamycin treatment for 24 hours, respectively. The ICso values of oxymatrine at 6, 12, 24, 48 and 72 hours were 27.27, 26.81, 24.75, 20.29 and 17.01 mg/mL, respectively. Conclusion Oxymatrine have bacteriostasis activity against Escherichia coli in vitro, and the the bacteriostasis effect enhance as the time went on, but the bacteriostasis activity is low.
