归芍滴丸的制备及其对东莨菪碱诱发的大鼠学习记忆障碍的改善 被引量:1

摘要 优化归芍滴丸的成型工艺,研究其对大鼠学习记忆障碍的影响。方法:采用正交设计方法,以外观质量、滴丸圆整度、溶散时限等为考察指标,以基质药物配比、药液温度、冷却柱长、冷凝液温度为实验因素,优选滴丸滴制条件;采用跳台实验,研究归芍滴丸对东莨菪碱所致大鼠学习记忆障碍的影响。结果:优化的归芍滴丸滴制工艺为:选用PEG 4000为基质,甲基硅油为冷凝剂,药物与基质配比为1∶2,药液温度为80℃,冷凝剂温度为10℃,滴速为30~40 d/min,滴距为3 cm,冷却柱长为15 cm。所制备的归芍滴丸平均丸重约50 mg/粒,有效成分阿魏酸和芍药苷的含量分别为0.27 mg/粒和0.24 mg/粒;与对照组和散剂组相比,归芍滴丸组大鼠的错误次数显著减少(P<0.001),潜伏期明显延长(P<0.001)。结论:本实验优化出的归芍滴丸滴制条件可行,归芍滴丸能显著促进小鼠的学习记忆能力的提升。 To optimize the preparing technology of Guishao Dropping Pills and study the effects of Guishao Dropping Pills on Scop-induced learning and memory impairment of mice. Methods: Orthogonal experiment was used to optimize the preparing technology of Guishao Dropping Pills as guided by the appearance quality, the dissolved time, the roundness of Guishao Dropping Pills. The step-down test for mice was adopted to study theeffects of Guishao Dropping Pills on Scop-induced learning and memory impairment. Results: The optimized preparing technology for Guishao Dropping Pills is: PEG 4000 as matrix, methyl-silicon oil as cooling agent, 1:2 for the ratio of drug to matrix, 80℃ for the temperature of drug solution, 15 cm for the length of cooling tube, 10℃ for the temperature of cooling agent, 3 cm for the dropping distance, 30 - 40 d. min^-1 for dropping speed. The weight of each pill is about 50rag and the contents of Ferulic acid and Paeoniflorin were 0. 27 mg and 0.24 mg for each pill, respectively. As compared with control group and powder group, the error frequency of experiment groups in the step-down test decreased significantly(P 〈0.001) , and the retention time was longer.
