摘要
目的建立顶空毛细管气相色谱法定量检测替加环素中的有机溶剂。方法采用气相色谱法测定。色谱柱:PEG-20M(30m×0.53mm,1μm);柱温采用程序升温;载气为氮气;采用氢离子火焰检测器。结果乙醚、甲醇、二氯甲烷和异丙醇均能得到有效分离;测得各组分的线性关系良好(相关系数均在0.999以上);平均回收率分别为99.4%,99.3%,97.5%和98.8%;最低检测限分别为0.42,0.61,1.53和0.65μg.mL-1。结论该方法灵敏,准确可靠,可用于替加环素中残留溶剂的检测。
Objective To establish a method of headspace capillary GC for the determination of residual solvents in tigecycline. Meth- ods The GC method was conducted on a PEG-20M capillary column(30 mX 0.53 mm, 1 btm) with procedural column temperature control. Nitrogen was used as the carrier gas and FID as the detector. Results The four residual solvents (diethyl ether,methanol, dichloromethane,and isopropanol) were completely separated,and the linearities were fairly good( r〉0. 999 ). The recoveries were 99.4 % ,99.3 %, 97.5 %, and 98.8 %, respectively. The limits of detection were 0. 42,0. 61,1. 53 and 0.65 μg · mL-1 , respectively. Conclusion The established method is sensitive and accurate, and can be used for the determination of residual solvents in tigecycline.
