摘要
目的观察川芎嗪对大鼠急性脊髓损伤模型损伤段NF-κB及I-κBα表达的影响。方法 SD大鼠110只,随机分为正常对照组、单纯脊髓损伤组和川芎嗪处理组。采用改良ALLEN氏打击法建立大鼠急性脊髓损伤模型。采用改良Rivlin斜板实验、BBB评分对大鼠脊髓功能进行行为学评分。在建模后1、3、6、12 h,1、3、5、7、14和21 d获取损伤段脊髓标本,HE染色观察其组织病理变化;免疫组织化学染色检测NF-κB及I-κBα表达。结果随着时间推移,术后斜板临界度数和BBB评分值均逐渐升高,CBS评分值逐渐降低,且术后7、14和21 d川芎嗪处理组斜板临界角度数均较单纯脊髓损伤组高(P<0.05),术后5、7、14和21 d实验组BBB评分值均较单纯脊髓损伤组高(P<0.05),而术后5、7、14和21 d,川芎嗪处理组CBS评分值均较对照组低(P<0.05)。术后1、3、5和7 d,川芎嗪处理组NF-κB阳性细胞表达数较单纯脊髓损伤组低(P<0.05),I-κBα阳性细胞表达数较单纯脊髓损伤组高(P<0.05)。结论川芎嗪能够促进大鼠急性继发性损伤后脊髓组织中I-κBα的表达,抑制NF-κB的表达,从而起到促进脊髓功能恢复的作用。
Objective To study the effect of Teramethylpyrazine(TMP) on expression of NF-κB and I-κBα in acute spinal cord injury(ASCI) in rats.Methods Allen'S weight drop method was used to establish acute spinal cord injury(SCI) rat model at T10 section and was conducted in 110 adult Sprague Dawley rats that were subsequently divided into sham operation group(group S,n=10);control group(group C,n=50);Tramethylpyrazine group(group T,n=50).The function of spinal cord was evaluated with animal behavioral scores by measuring modified Rivilin loxotic plate test and counting BBB and CBS score at 1,3,5,7,14 and 21 days postoperatively.The injured spinal cord samples were harvested at 1,3,6,12 hours,1,3,5,7,14 and 21 days postoperatively(n=5 for each time point) for the preparation of continuous histological sections that were analyzed the expression of NF-κB and I-κBα by immunohistochemistry method.Results Group T had significantly higher degrees than group C at 7,14 and 21 days in modified Rivlin loxotic plate test after SCI(P0.05);Group T had significantly higher scores in BBB score than group C at 5,7,14 and 21 days after SCI respectively(P0.05).Group C had significantly higher scores in CBS score than group T at 5,7,14 and 21 days after SCI respectively(P0.05).The number of NF-κB positive cells were significantly lower in group T than group C at 1,3,5 and 7 days after SCI(P0.05);The number of I-κBα positive cells were significantly lower in group C than group T at 1,3,5 and 7 days after SCI(P0.05).Conclusions The TMP can promote repairing of injured spinal cord tissue through improving the I-κBα expression and decreasing the NF-κB expressions in cells after ASCI.
