摘要
目的探讨上调缺氧诱导因子-1(HIF-1)对大鼠球囊损伤后内膜增生的影响。方法 30只雄性健康SD大鼠随机分为假手术组、模型组和干预组,模型组和干预组均采用1.5F球囊导管建立颈动脉球囊损伤模型。于术后第1 d开始,干预组给予含羞草素(L-Mim)100 mg/(kg.d)灌胃,假手术组和模型组给予等量生理盐水灌胃。干预14 d后取损伤血管段,测定内膜增生情况,并用Western blotting法检测HIF-1的蛋白表达。结果术后14 d,模型组和干预组均发生明显内膜增生,内膜面积和内膜与中膜面积比均显著高于假手术组(P<0.01),而干预组上述指标均低于模型组(P<0.01)。与模型组比较,干预组损伤血管中HIF-1的蛋白表达水平显著升高(P<0.05)。结论采用L-Mim上调HIF-1可显著削弱球囊损伤导致的内膜增生。
Objective To investigate the effects of up-regulating hypoxia-inducible factor-1(HIF-1)on intimal hyperplasia after saccule injury in rats.Methods Thirty healthy male SD rats were randomly divided into sham operation group,model group and intervention group.The rat model of carotid artery saccule injury was established using 1.5 F saccule catheter in the model group and the intervention group.From the first day after the operation,the intervention group received intragastric administration with L-mimosine(L-Mim)100 mg/(kg·d).The sham operation group and the model group received intragastric administration with the same dosage of saline.After intervention for 14 d,the injured section of the blood vessel was taken for the detection of intimal hyperplasia,and Western blotting was used to determine the protein expression of HIF-1.Results Obvious intimal hyperplasia was observed in both model and intervention group 14 d after the operation.The area of intima and the ratio of intimal area to tunica media area in the two groups were all significantly higher than those in the sham operation group(P〈0.01).The indexes above in the intervention group were all lower than those in the model group(P〈0.01).Compared with the model group,the protein expression of HIF-1 in the injured blood vessel of the intervention group significantly increased(P〈0.05).Conclusion L-Mim may significantly attenuate intimal hyperplasia caused by saccule injury through up-regulating HIF-1 expression.
