摘要
目的研究高压氧(hyperbaricoxygen,HBO)对维生素C药代动力学的影响。方法采用数字表法将240只小鼠随机分为2组,每组120只,2组均按3g/kg剂量单次灌服维生素C注射液。HBO组灌服后立即进入高压氧舱,稳压(0.10MPa)吸纯氧20min,升压、减压各5min;对照组仅单次灌服维生素C注射液。2组在给药前及给药后不同时间点各取10只小鼠,摘眼珠取血,采用高效液相色谱法测定血浆中维生素C的浓度,计算药代动力学参数。结果HBO组赤池信息量准则(AIC)、血浆半衰期(t1/2)、达峰时间(tpeak)、峰浓度(cmax)、平均血药浓度-时间曲线下面积(AUC)分别为-26.4、5.4h、0.8h、17.9mg/L、126.2(mg·h)/L;对照组AIC、t1/2、tpeak、cmax、AUC分别为-3.8、11.6h、1.1h、11.2mg/L、155.5(mg·h)/L。结论HBO可以明显加快小鼠体内维生素C的吸收,对药代动力学参数有明显的影响。
Objective To investigate the effects of hyperbaric oxygen (HBO) on the pharmacokinetics of vitamin C: Methods Two hundred and forty mice were randomly divided into 2 groups, the HBO group and the control group, each consisting of 120 animals. The 2 groups of animals received gavage of vitamin C with a single-close of 3g/kg. The animals in the HBO group were immediately put into the hyperbaric oxygen chamber and maintained at a pressure of 0. 1 MPa for a duration of 20 min. The time used for compression and decompression was all 5 rain. The animals in the control group were just given a single gavage of vitamin C. Ten animals from each group were chosen at different time points both before and after drug administration, for collection of blood samples from the eyeballs. Concentrations of vitamin C in serum were monitored with high performance liquid chromatography, and pharmacokinetic indices were calculated accordingly. Results The main pharmaeokinetic parameters of the HBO group were as follows : AIC ( - 26.40 ) , t1/2 ( 5.4 h) , Tpeak (0.8 h), Cmax( 17.9 rag/L), AUC[ 126.2( mg ·h)/L], while the corresponding results of the control group were AIC( -3.8), t1/2(11.6h), Tpeak(1.1 h), Cmax(11.2 m/L), AUC[ 155.5(mg ·h)/L]. Conclusions HBO could obviously accelerate the absorption of vitamin C and seemed to have significant effect on pharmacokinetic parameters.
