摘要
目的:给予妊娠期C57BL/6母鼠不同剂量的微量元素锌,探讨母体锌含量与其子代腭裂畸形发生之间的关系以及锌对致腭裂环境因素的拮抗作用。方法:将C57BL/6孕鼠随机分为Ⅰ和Ⅱ两大组,Ⅰ组为单纯锌干预组,即正常对照组、高锌组(a组)、低锌组(b组),Ⅱ组为锌+外界因素干预组,即高温组(A组),高温+高锌组(A’组),内毒素组(B组),内毒素+高锌组(B’组),并给予相应干预,于胚胎18d处死孕鼠,观察胚胎腭裂发生情况。结果:对照组和实验a、b、A、A’、B、B’组胎鼠腭裂发生率分别为3.13%、2.94%、27.27%、21.21%、5.02%、27.59%、6.06%。实验a组和对照组间胚胎腭裂率比较,无统计学意义,二者与实验b组间比较(P<0.05);实验A’、B’组胎鼠的腭裂发生率与对照组比较,无统计学意义;实验A和A’组间以及实验B和B’组间胎鼠腭裂发生率比较(P<0.05),有统计学意义。结论:母体血清锌含量与子代腭裂的发生有密切关系,血清中锌的含量过低易致腭裂的发生,在相同的致畸条件下,通过饮食摄入足量的微量元素锌可以对腭裂畸形的产生起到一定的预防作用。
Objective: To study the relations between the content of microelement zinc in mother and the occurrence of cleft palate in fetus in order to provide some theories for preventing occurrence of cleft palate. Methods: The fertiiization mice of C57BL/6 were divided into two group at the first day. Group Ⅰ was subdivided into the control group , the high zinc food group(a), the low zinc food group(b); group Ⅱ was subdivided into A group, A' group, B group and B group, in which were treated with high temperature, high temperature+ high zinc food, injection endotoxin group and injection endotoxin + high zinc food respectively. Dams were killed and fetus were collected at El8. We observed and recorded the number of mouse embryonic cleft palate. Results: There is no statistical significance in occurrence rate of mouse embryonic cleft palate between the control group and A group; there is statistical significance between the control group or A group and B group (P〈0.05). Occurrence rate of mouse em- bryonic cleft palate between the control group and A',B' group have no significant deviation, but there is statistical significance between A'/B' group and A/B group (P〈0.05). Conclusion: The low content of the serum zinc in parent could increase the risk of cleft palate in fetus. Furthermore, high content of the serum zinc in parent could decrease the risk of cleft palate in fetus when the dams were affected by environmental adverse factors during pregnancy.
