丙戊酸钠抑制口腔鳞状细胞癌生长的实验研究

摘要 目的:本实验探讨组蛋白去乙酰化酶抑制剂丙戊酸钠(VPA)对口腔鳞状细胞癌(Tca-8113)生长抑制情况以及促进凋亡的作用。方法:采用0、0.1、2.5、5、10、12.5、25、50、100mmol/L丙戊酸钠对口腔鳞状细胞癌进行干预,倒置相差显微镜观察细胞形态改变;CCK-8法检测细胞增殖活性;ELISA方法及Annexin-V-FITC/PI双染法检测对细胞凋亡的作用。结果:倒置相差显微镜下观察到对照组细胞呈多边形,贴壁,生长活跃;实验组细胞形态改变明显,大部分细胞核固缩,细胞变小、变圆、脱壁。细胞生长曲线显示,随丙戊酸钠浓度增加、时间延长,Tca-8113细胞生长明显受到抑制,各实验组细胞存活率与对照组相比(P<0.05),差别有统计学意义。2.5、10mmol/L丙戊酸钠作用24h,细胞总凋亡率分别为9.2%和26.7%,与对照组3.6%相比,差别均有统计学意义。结论:丙戊酸钠体外抑制口腔鳞状细胞癌生长,呈时间-剂量依赖性,并诱导细胞凋亡。 Objective: To investigate the effects of valproic acid on the proliferation of oral squamous cell carcinoma (Tca-8113) and promoting apoptosis. Methods: 0, 0. 1, 2.5, 5, 10, 12. 5, 25, 50, 100 mmol/L valproic acid were intervented on oral squamous cell carcinoma. The morphologic changes of Tca-8113 cells were observed by the inverted phase contrastmicroscope. The cell proliferative activity was measured by CCK-8 assay, the cell apototic rates were detected by flow cytometry through Annexin-V-FITC/PI staining and ELISA. Results: Under the inverted phase contrast microscope, no changes of cell morphology could be observed in the control, cells grow polygonal, adherent and actively. But the changes could be observed in groups treated with va|proic acid at 10, 25mmol/L for 24h. The cells became smaller and rounder, and shrank cells increased. A time and dose-dependent inhibition was detected in Tca-8113 cells that were treated with different concentration of valproic acid for 12, 24, 48 and 72h. And there was difference of proliferative inhibition between the control and the treated groups (P〈0. 05). Treated by 2.5, 10mmol/L valproic acid for 24h, cell apoptosis rate was 9.2% and 26.70//oo. The difference was statistically significant. Conclusion: Valproic acid inhibits the growth of oral squamous cell carcinoma, which was time-dose dependent, and can promote apoptosis.
