摘要
目的观察豆根管食通口服液对食管癌模型大鼠细胞凋亡及免疫逃逸的影响,从分子生物学角度分析其作用机制。方法健康Wistar大鼠随机选择15只为空白组,其余大鼠均腹部皮下注射甲基戊基亚硝胺5mg/kg建立食管癌模型大鼠,将造模成功的75只大鼠随机分为模型组、高剂量组、中剂量组、低剂量组、替加氟组各15只,低、中、高剂量组分别按5g/kg、10g/kg、15g/kg给予豆根管食通口服液,空白组、模型组给等予容积生理盐水,替加氟组给予替加氟片20mg/kg,各组均于造模成功24h后灌胃给药,每日1次,连续4周。末次给药24h后取瘤检测,原位末端标记法检测细胞凋亡,免疫组织化学法检测Fas、FasL蛋白表达。结果高、中、低剂量组及替加氟组食管癌细胞的凋亡指数与空白组、模型组比较差异均有统计学意义(P<0.05),并且高剂量组明显优于替加氟组、低剂量组(P<0.05)。模型组Fas蛋白表达阳性率低于空白组、FasL蛋白表达高于空白组(P<0.05或P<0.01);各治疗组Fas蛋白表达阳性率均高于模型组(P<0.05),高剂量组Fas蛋白表达阳性率高于替加氟组和低剂量组(P<0.05);高、中、低剂量组FasL蛋白表达阳性率明显低于模型组(P<0.05或P<0.01)。结论豆根管食通口服液能促进模型大鼠食管癌细胞凋亡,其作用机制可能与其提高Fas表达、降低FasL表达有关,并且存在一定的量效关系,高剂量的豆根管食通口服液作用优于替加氟片。
Objective To observe the effect of Dougen Guanshitong Oral Liquid [oral liquid of Dougen (Radix et Rhizoma Sophorae Tonkinensis) for esophageal cancer] on apoptosis and immune evasion in rats with esophageal cancer and discuss its mechanism based on molecular biology. Methods Fifteen health Wistar rats were selected into the control group. The esopha- geal cancer model was established by abdominal subcutaneous injection of methyl-n-amylnitrosamine (5mg/kg). The model rats were randomized into the model group, large-dose group, middle-dose group, small-dose group and tegafur group, with 15 in each. The small-dose group, middle-dose group and large-dose group were given 5g/kg, 10g/kg and 15g/kg of Dougen Guan- shitong Oral Liquid respectively. The control group and model group were given normal saline of equal amount. The tegafur group was given 20mg/kg of tegafur tablet. Administration began at the 24th after model establishment through gastric feed- ing, once a day, 4 weeks in all. Twenty-four hours after the last administration, the tumors were removed. The apoptosis was detected by terminal in situ nick end labeling. The immunohistoehemistry method was used to detect the expression of Fas and FasL protein. Results Comparing with the control group and model group, the apoptotic index in the large-dose group, mid- dle-dose group, small-dose group and tegafur group was statistically different (P^0.05). The effect in the large-dose group was significantly superior to those in the tegafur group and small-dose group (P%0.05). Comparing with the control group, the positive rate of Fas expression in the model group was significantly decreased, but the positive rate of FasL expression was significantly increased (P^0.05 or P%0.01). The positive rate of Fas expression in all the treatment groups was significantly higher than that in the model group (P^0.05). The positive rate of Fas expression in the large-dose group was significantly higher than those in the tegafur group and small-dose group (P〈0.05). Comparing with the model group, the positive rate of FasL expression in the large-dose group, middle-dose group and small-dose group was significantly decreased (P〈0.05 or P〈0.01). Conclusion Dougen Guanshitong Oral Liquid could promote the apoptosis of esophageal cancer in a dose-effect manner and its mechanism maybe relevant to increasing Fas expression and decreasing FasL expression. The effect of lage-dose of Dougen Guanshitong Oral Liquid is superior to that of tegafur tablet.
