胃癌组织中半胱氨酸蛋白酶-8的表达及临床意义 被引量:2

The expression of Caspase-8 in gastric carcinoma tissues and its clinical significance

摘要 目的研究胃癌组织中半胱氨酸蛋白酶-8(Gaspase-8)的表达特点及其与临床病理学特征的关系。方法采用逆转录聚合酶链反应法及免疫组化法检测54例胃癌及其癌周组织中Caspase.8mRNA及蛋白的表达。结果胃癌组织中Caspase-8mRNA表达明显低于癌周组织(0.154±0.065vs0.3944-0.107,t=14.04,P〈0.01),与淋巴结转移、肿瘤分期、分化程度有关(P〈0.01);胃癌组织中Caspase-8蛋白的阳性率也明显低于癌周组织(31.48%vs85.19%,X^2=32.04,P〈0.01),与肿瘤分期、分化程度有关(P〈0.05)。结论胃癌组织中Caspase-8表达下调,可能与胃癌发生发展密切相关。 Objective To investigate the expression characteristics of Caspase-8 in gastric carcino- ma tissues and its correlation with clinical pathological features. Methods The mRNA and protein of Caspase-8 in gastric carcinoma and its surrounding tissues were detected by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry, respectively. Results The Caspase-8 mRNA in gas- trie carcinoma tissues was obviously lower than that in its surrounding ones (0. 154 ± 0. 065 vs 0. 394 ±0. 107, t = 14. 04, P 〈 0.01 ), which was correlated with lymph node metastasis, tumor stage, differentia- tion grade ( P 〈0. 01 ). The Caspase-8 protein in gastric carcinoma tissues was also significantly lower than that in its surrounding ones (31.48% vs 85.19% , X^2 = 32.04, P 〈 0. 01 ), which was correlated with tumor stage, differentiation grade ( P 〈 0. 05). Conclusioas Caspase-8 is low-expressed in gastric carci- noma, which may play an important role in the occurrence and progression of gastric carcinoma.
