小鼠哮喘死亡主支气管上皮LCR值及mmp-2的表达

An experimental study on LCR and expression of mmp-2 in the epithelial cells of the primary bronchus in the case of death due to asthma in mice

摘要 目的观察基质金属蛋白酶2(mmp-2)在实验性小鼠支气管哮喘死亡模型主支气管上皮的表达规律。方法 6~8周雄性健康小鼠30只,随机取20只建立哮喘死亡模型(B组),10只为空白对照组(A组)。提取小鼠死后主支气管组织,经HE和免疫组化染色(SP法)后,用图像分析系统分别观测并测量小鼠支气管软骨厚度和黏膜固有层厚度比值(LCR),并用免疫蛋白印记法(Western blot)检测主支气管上皮mmp-2。实验所得数据用SPSS 11.0统计学软件进行分析。结果 B组主支气管LCR 1~3周内逐渐上升,mmp-2免疫组化阳性染色平均光密度值(MOD)逐渐升高、灰度值亦升高,与A组比较,两组数据差异均具有显著性意义(P<0.01)。结论 LCR可反映气道壁的增厚和间质纤维化,并结合mmp-2灰度值增高变化,可望成为诊断哮喘猝死的辅助指标之一。 Objective To observe the expression of matrix metalloproteinas-2(mmp-2) in the epithelial cells of the primary bronchus in deaths from due to bronchial asthma in mice.Methods 30 male Balb/c mice of 6-8 weeks were divided into control group(group A,n=10)and deaths due to bronchial asthma group(group B,n=20).The thickness of the lamina propria mucosae and bronchial cartilage was measured,and ratio of the lamina propria mucosae to bronchial cartilage(LCR) was calculated.Immunohistochemistry SP and Western blot were used to detect the expression of mmp-2 in the epithelial cells of the primary bronchus.Computerized images analysis and the expressive differences in the two groups were compared by statistics analysis.Results The LCR was obviously piecemeal increased in group B 1-3 weeks after onset of asthma.The mean optical density(MOD)of mmp-2 product was significantly increased.There were significant difference between group B and group A(P〈0.01).The gray value for mmp-2 was significantly higher in group B than that in group A(P〈0.01).Conclusion LCR may predict increased interstitial fibrosis and thickness of tracheal wall.,which is expected to be a supplimetal indicator for diagnosis of sudden death due to bronchial asthma in combination with increased gray value of mmp-2.
