SHH蛋白在大肠腺瘤和大肠癌中的表达及意义 被引量:1

Expressions and Significance of Sonic Hedgehog in Adenoma and Colorectal Carcinoma

摘要 目的:探讨Shh(sonic hedgehog)在大肠腺瘤及大肠癌中的表达及其临床意义。方法:应用免疫组织化学方法检测48例大肠腺瘤及50例大肠癌中Shh蛋白的表达,并与临床病理因素进行相关性分析。结果:Shh蛋白的阳性信号分布在大肠腺瘤和大肠癌细胞质内,在大肠腺瘤和大肠癌中的阳性表达率分别为64.6%(31/48)和58.0%(29/50),明显高于正常组织的20.0%(5/25)(P<0.01,P<0.05);在大肠腺瘤中,Shh的染色评分随腺瘤组织学分级的增加而呈显著性升高(P<0.05)。Shh在早期大肠癌的阳性表达率和染色评分都高于进展期大肠癌,差异均有统计学意义(P<0.01和P<0.0001)。Shh在大肠腺瘤和大肠癌中的表达与性别、年龄、肿瘤部位和大小及组织学类型无关(P>0.05)。结论:Shh信号通路的异常激活在大肠癌发生过程中发挥一定的作用,Shh可作为大肠癌早期诊断的生物学指标及潜在的治疗靶点。 Objective :To explore the expressions and significance of Shh (sonic hedgehog)in colorectal adenoma and colorectal carcinoma (CRC). Methods:The expressions of Shh in 48 cases with colorectal adenomas and 50 cases with CRCs were detected and evaluated by immunohistochemistry, and its correlation with clinicopathological factors was statis- tically analyzed to investigate. Results : Shh protein was detected in the cytoplasm of colorectal adenoma and CRC cells, the positive rate of Shh was in colorectal adenoma and CRC were 64.6% (31/48) and 58.0% (29/50)respectively ,which were significantly higher than that in normal tissue 20.0% (5/25) (P 〈 0.01, P 〈 O. 05 ) ;For staining score evaluation, the expression of Shh in colorectal adenoma were significantly stronger with the increase in histological grade ( P 〈 0.05 ). Both Shh expressions and staining score evaluation in initial CRCs were significantly higher than that in advanced ones( P 〈0. 0I, P 〈0.0001 ). The expression of Shh in the colorectal adenoma and CRC had no correlation with Sex ,age ,tumor position and size,and pathologic type ( P 〉 0.05 ). Conclusion:The aberrant activation of Shh signaling pathway plays some roles in progression of CRC. Shh may be indicators for early diagnoses of CRC and potential therapeutic targets.
