干扰素-γ增强RAW264.7巨噬细胞对阿萨希毛孢子菌杀伤作用的机制研究 被引量:5

摘要 目的探讨干扰素-γ(IFN-γ)对RAW264.7巨噬细胞(Macrophage,Mφ)体外抗阿萨希毛孢子菌(Trichosporon asahii,T.asahii)杀伤作用的机制。方法不同浓度(10、100、1000U/ml)的IFN-γ与RAW264.7共同培养,再加入阿萨希毛孢子菌培养后检测菌落形成单位(CFU/ml),计算各生长抑制率。在上述体系上清液中分别检测亚硝酸根离子(NO2-)。将巨噬细胞与不同终浓度的IFN-γ共同孵育后,分别检测超氧阴离子(O2-)的OD值。结果 IFN-γ浓度分别为10、100、1000U/ml时,T.asahii的生长抑制率分别为25.21%、41.95%、55.83%,组间差异具有显著统计学意义(P<0.01);上清液中NO2-的浓度分别为89.16±3.32、108.07±3.81、126.44±3.52,与空白对照组(74.47±1.48)相比差异具有显著性意义(P<0.01);上清液中O2-的OD值分别为0.317±0.006、0.424±0.006、0.518±0.006,与对照组相比差异具有显著性意义(P<0.01)。结论随着IFN-γ浓度的增加,T.asahii生长抑制率增加。IFN-γ通过增强Mφ产生一氧化氮(NO)及超氧阴离子(O2-)来发挥体外抗T.asahii的杀伤作用。 Objective To investigate the effect of IFN-γ on the killing mechanism of RAW264.7 macrophage against Trichosporon asahii(T.asahii) in vitro.Methods The various concentrations(10,100,1000U/ml) of interferon-γ with RAW 264.7 were incubated,and then cultured with T.asahii to assess the killing activity by counting the number of colony forming units per milliliter(CFU/ml) of T.asahii.Then calculate the T.asahii growth inhibition rate,and measure Nitrite ion(NO2-) in the supernatant respectively.Each well added by the above different terminal concentrations of IFN-γ incubated with RAW264.7 to determine the superoxide anion(O2-) respectively in the supernatant.Results With the concentration of 10,100 and 1000U/ml of IFN-γ,the growth inhibition rates of T.asahii were 25.21%,41.95%,55.83% respectively.There was significant difference between each group(P0.01).In the supernatant the concentrations of Nitrite ion were 89.16±3.32,108.07±3.81,126.44±3.52 respectively.There were significant difference(P0.01) compared to the blank control group(74.47±1.48).With the concentrations of IFN-γ rising(10,100,1000U/ml respectively),the OD value of the O2-measured in the supernatant were increasing(0.317±0.006、0.424±0.006,0.518±0.006 respectively).And the blank control group was 0.222±0.008,there was significant difference between each group(P0.01).Conclusion With the concentrations of IFN-γ rising,the growth inhibition rates of T.asahii are increasing.IFN-γ plays a role in killing effect of T.asahii in vitro by enhancing macrophage producing nitrogen monoxidum(NO) and superoxide anion(O2-).
