摘要
目的 探讨 ATP生物发光法作为化疗药物敏感性预测方法的可行性。方法 参照 Sevin等 [1 ] 的 2 4孔培养板培养、手工操作检测的方法 ,改进成 96孔微量培养板全自动检测法。通过检测 K 5 6 2细胞对 11种常见抗癌药物的敏感性 ,并与 MTT比色法比较。结果 两者有良好的相关性 (r= 0 .885 ) ,但 ATP法所测得的变异系数明显小于 MTT法(前者为 1.4%~ 13.6 % ,后者为 0~ 2 4.2 % ,P=0 .0 11)。以 ATP标准品作标准曲线 ,当 ATP的浓度在 10 - 6~ 10 - 1 2范围内时 ,ATP的浓度的对数与发光测定值的对数之间有较好的线性关系。ATP测定值与 K5 6 2细胞实际数之间相关性亦很好 (r=0 .994)。结论 ATP生物发光法是一种敏感、可靠的药敏实验方法 。
Objective To explore the feasibility of an adenosine triphosphate (ATP) bioluminescence assay for chemosensitivity test of cancer.Methods Eleven anticancer drugs were tested using the ATP assay on K562 cell line compared with the diphenyltetrazolium bromide (MTT) method. Results The results of ATP bioluminescent assay correlated well with those of the MTT assay ( r= 0 885), the coefficient variation (CV) of measurement for each drug concentration ranged from 1 4%-13 6% with the ATP method, compared with 0%-24 2% of the MTT method ( P =0 011). A linear relationship obtained between the log luminescence and the log concentration of ATP in the range of 10 -6 ~10 -12 mol ATP concentration.Conclusion The ATP bioluminescence assay is an accurate and sensitive in vitro test for measuring the sensitivity of anticancer agents, and deserves clinical application.
