摘要
通过测定保加利亚乳杆菌在以胰蛋白胨为主要氮源和在以大豆蛋白胨为主要氮源的培养基中培养不同时间后的光密度值(OD)、活菌数、湿菌泥得率和培养过程中细胞革兰氏染色的形态,来分析两种培养条件下的差异。结果表明:保加利亚乳杆菌在以胰蛋白胨为主要氮源的培养基中培养后测得的OD590 nm值要比在以大豆蛋白胨为主要氮源的培养基中的高,活菌数也较多,但由于其细胞形态相对比较细小,因此所获得的湿菌泥得率反而相对比较少。该结果对今后工业化生产该菌种直投式发酵剂的氮源选取提供有利参考,同时也进一步阐明了保加利亚乳杆菌在不同氮源培养基中生长的不同特性。
The growth characteristics of Lactobacillus bulgaricus differ with different nitrogen sources.This paper describes the differences in optimal density(OD590nm),viable cell count,the yield of wet cell paste,and Gram staining morphology during the cultivation of Lactobacillus bulgaricus in different media with tryptone or soya peptone as the main nitrogen source.Cultivation in the medium with the main nitrogen source tryptone provided higher OD590nm and more viable cells.However,the yield of wet cell paste was lower than that obtained using soya peptone as the main nitrogen as a result of the smaller morphology.These findings could provide useful references for the industrial production of DVS starter culture from Lactobacillus bulgaricus and further elucidation of different growth characteristics of the strain in media with different nitrogen sources.
