摘要
小麦高分子量麦谷蛋白亚基(HMW-GS)组成与其加工品质密切相关,建立其相关简便有效的遴选体系对小麦品质评价和优质品种选育具有重要意义。本研究选用对小麦品质有重要影响的HMW-GS基因的特异性分子标记,基于26份已知HMW-GS组成的小麦品种(系)检测验证基础上,建立强筋小麦HMW-GS基因的多重PCR检测体系。已建立了4套多重PCR体系:体系Ⅰ同时检测Axnull与By8基因,体系Ⅱ检测By8与Dx5基因,体系Ⅲ同时检测Axnull、By8与Dx5基因,体系Ⅳ同时检测Axnull、Bx14与Dx5基因。利用每个体系对26份品种(系)进行检测,其结果与已知结果一致,说明创建的4套多重PCR体系稳定结果可靠,可用于强筋小麦品种的分子辅助选择和品质评价。
High-molecular-weight glutenin subunits(HMW-GS) are closely correlated with the processing quality of common wheat.Establishment related select systems which are simple and convenient is very important to evaluate wheat quality and select new wheat cultivars with good qualities.According to the molecular markers for genes controlling important qualities,four types of multiplex PCR detection systems for strong-gluten wheat were developed and validated using 26 Chinese wheat cultivars and advanced lines with known HMW-GS(genes).The first multiplex PCR was used to simultaneously detect genes Axnull and By8,the second one was to detect the genes By8 and Dx5,the third multiplex PCR included three genes Axnull,By8 and Dx5,the fourth one was to detect the genes Axnull,Bx14 and Dx5.After the 26 Chinese wheat cultivars were detected using the four types of multiplex PCRs,respectively,their results were consistent with those of the known.The results indicated that the four multiplex PCRs established in the present study were stable and reliable.The multiplex PCRs would be effectively used to evaluate and select wheat quality properties in wheat breeding programs.
