摘要
明确来源于野生二粒小麦的抗白粉病基因PmAS846所在的染色体臂。采用端体和SSR标记对普通小麦种质N9134携带的PmAS846基因进行了遗传分析。中国春(Chinese Spring,CS)、CS5BL双端体系与N9134杂交F1高抗白粉病,CS/N9134的F2和CS5BL双端体系/N9134//CS BC1F1抗病植株和感病植株比例分别符合期望比例3∶1和1∶1。CS 5BL双端体系/N9134//CS BC1F1 64个植株的根尖染色体核型和白粉病抗性鉴定发现,12个植株在5B染色体着丝点和抗性基因间发生了交换。PmAS846的SSR标记Xgwm67在CS第五部分同源群的缺体-四体系和双端体系的扩增,该标记在CSN5BT5D系中缺失,而在CSDt5BL系中存在。结果表明:PmAS846位于小麦5B染色体长臂上,该基因距5B染色体着丝点18.75个交换单位。
A powdery mildew resistance gene PmAS846 derived from Triticum dicoccoides accession As846 and was located on 5B chromosome.The aim of this research was to locate this gene to chromosome arm.Genetic analysis of PmAS846 transferred to common wheat line N9134 was done by ditelocentric line and SSR markers.F1 progeny between common wheat CS(Chinese Spring)disomics,CSDt5BL(CS 5BL ditelocentric line)and N9134 were highly resistant to powdery mildew.The ratio of the resistant and susceptible plants in CS/N9134 F2 progeny and CSDt5BL/N9134//CS BC1F1 progeny fitted the expected 3 ∶ 1 and 1 ∶ 1,respectively.The karyotype analysis of root tip cell and powdery mildew resistance for 64 CS5BL/N9134//CS BC1F1 plants was done,and the crossover types had frequencies of 12.The analysis results of SSR marker Xgwm67 in CSN5AT5D,CSN5BT5D,CSN5DT5A,CSDt5AL,CSDt5BL and CSDt5DL indicated that the marker was absent in CSN5BT5D and present in CS Dt5BL.The results showed that PmAS846 was located on 5BL and the distance of PmAS846 and the 5B centromere was 18.75 map units.
