金华猪DGAT1、DGAT2基因发育性表达特点及其与肌内脂肪的关联分析 被引量:9

Developmental Expression Characteristics of DGAT1 and DGAT2 Gene and Its Association Analysis with Intramuscular Fat in Jinhua Pig

摘要 二脂酰甘油酰基转移酶是催化甘油三酯合成最后一步反应的关键酶,包括DGAT1和DGAT2两种;阐明其在发育过程中的表达规律,对于找到控制猪脂肪沉积能力的基因十分必要。运用实时荧光定量RT-PCR的方法,对60,90,120日龄金华猪肝脏、皮脂和眼肉中DGAT的mRNA表达量进行了分析,同时分析了DGAT基因mRNA表达量与肌内脂肪的相关性。结果表明,金华猪DGAT1、DGAT2基因在不同日龄的表达差异均不显著(P>0.05);在肝脏、皮脂和眼肉中,DGAT1基因表达差异均极显著(P<0.01);皮脂与肝脏、眼肉的DGAT2基因表达差异均极显著(P<0.01),而肝脏与眼肉的DGAT2基因表达差异不显著(P>0.05);而且各日龄均是皮脂中DGAT1、DGAT2基因表达量最高,肝脏次之,眼肉中最低。提示DGAT2基因在猪脂肪沉积中发挥着比DGAT1更重要的作用。研究还发现,DGAT1和DGAT2基因表达量与肌内脂肪含量无显著意义的相关(P>0.05)。 The final and critical step in the synthesis of triglycerides is catalyzed by two acyl-coA: diacylglycerol acyhransferase(DGAT)enzymes,i, e. DGAT1 and D GAT2. Analysis on the developmental expression of DGAT1 and DGAT2 mRNA in three tissues is a.prerequisite for the identification of genes underling fat deposition. The DGAT1 and DGAT'2 mRNA expression of Jinhua pig 60-day and 90-day and 120-day was investigated by Real-time fluorescent quantitative RT-PCR in this study,and analysis correlation of DGAT gene expression with IMF( intramuscular fat). The results showed there was not any different(P 〉 0. 05 ) expression of DGAT1 and DGAT2 in different days of the Jinhua pigs. There was significant difference(P 〈 0. O1 ) expression of DGAT1 in the different tissues. There was significant difference ( P 〈 0.01 ) between expression of DGAT2 in the SF and L/RE, whereas no any difference( P 〉0.05 )was observed between the L and RE. The lowest level of expression of DGAT1 and DGAT2 was found in the RE,higher in the L,and the highest in the SF. These results suggest that DGAT2 gene play a key important role in pig fat deposition than DGAT1 gene. Correlation between DGAT1 and DGAT2 gene expression with IMF content was not remarkable ( P 〉 0.05 ).
