摘要 目的:研究八角莲对大鼠肝组织超氧化物歧化酶(superoxide.dimutase,SOD)、丙二醛(MDA)、大鼠活性氧簇(ROS)、丙氨酸转氨酶(ALT)、天门冬氨酸氨基转移酶(AST)、γ-谷氨酰转移酶(γ-GT)及肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的影响,探讨八角莲致大鼠肝脏损害的毒性机制。方法:分别给予SD大鼠八角莲水提液与醇提液连续灌胃3 d后,取正常组及八角莲给药组(3.50和2.96 g·kg^(-1))大鼠肝组织,制成10%肝匀浆,按试剂盒方法测定肝组织中SOD、MDA、ROS活性和ALT、AST、γ-GT含量,计算SOD/MDA;采用双抗体夹心ABC-ELISA法测定大鼠肝组织TNF-α、IL-6含量。结果:与正常组比较,八角莲水提组与醇提组大鼠肝组织SOD下降,MDA上升,SOD/MDA下降,ROS值上升(P<0.05);ALT、AST、γ-GT、TNF-α、IL-6含量升高(P<0.05)。结论:大鼠灌胃给予八角莲后,可对肝脏产生明显的毒性,其机制可能与影响自由基及炎症因子有关。 Objective : To study the effects of dysosma veitchii on SOD, MDA, ROS, ALT, AST, γ-GT, TNF-α and IL-6 in rats to explore the toxicity mechanism of dysosma veitchii. Method: SD rats were administrated water extract and ethanol extract of dysosma veitchii (3.50 and 2. 96 g. kg-1 ) by garage for 3 days, respectively. The livers of the normal group and the two dysosma veitchii groups were withdrawn and homogenized to obtain 10% liver homogenate. The activity of SOD, MDA and ROS and the content of ALT, AST and γ-GT were determined by kit method. The ratio of SOD/MDA was calculated. ABC-double antibody sandwich ELISA determination was applied to obtain the contents of TNF-α and IL-6. Result: Compared with the normal group, the two dysosma veitchii groups showed decreased SOD and increased MDA resulting in decreased SOD/MDA. ROS values were increased and ALT, AST, γ-GT,TNF-α and IL-6 levels were increased as well (P 〈 0. 05 ). Conclusion : Dysosma veitchii shows significant liver toxicity in rats, which may be related with the effects on free radicals and inflammatory cytokines.
