摘要
为探索重组绿色木霉源双功能壳聚糖酶(rCCBE)在毕赤酵母中表达的最佳诱导条件,本研究在对甲醇浓度、诱导pH、诱导温度和诱导时间这4个因素进行单因素试验基础上,运用Box-Behnken设计的响应面试验对诱导条件进行了优化。根据回归分析确定了影响rCCBE表达的因子,求得最佳诱导条件为:诱导甲醇浓度为0.92%、pH为6.26,诱导温度为28.42℃、诱导时间80.85 h,此工艺条件下双功能酶的实测壳聚糖和CMC降解活力分别为7.32 kat/mL和7.15 kat/mL,比优化前提高了76.4%。回归模型的预测值与实测值的相对误差<1%,说明该回归方程与实际情况拟合很好。
In order to optimize the conditions of expressing recombinant Trichoderma viride bifunctional chitosanase-cellulase(rCCBE) induced by methanol,the response surface methodology designed by Box-Behnken was applied based on single factor tests.The factors relating to the expression of rCCBE was obtained with method of regression analysis.The optimum inducing conditions were as follows: methanol concentration of 0.92 %,temperature of 28.42 ℃ for 80.85 h,pH 6.26.Under the above conditions,the actual chitosanase and CMCase activities of rCCBE were 7.32 kat/mL and 7.15 kat/mL,respectively.The optimal activities ware 76.4 % higher than before.The relative error was1 % between actual chitosanase activity and the activity calculated according to the regression model,which suggested that regression equation fitted well with actual condition.
