摘要
目的 观察巨噬细胞调理液 (MCM)能否促进培养大鼠视网膜神经细胞 (RN)的存活和生长 ,以探讨其损伤后的修复问题。方法 将生后 2~ 3天Sprague Dawley大鼠视网膜组织经胰蛋白酶消化后进行培养 ,并设对照组。用MTT比色法测量存活细胞的OD值。结果 培养在鼠尾胶原上的RN生长良好 ,且部分细胞伸出突起。培养 1、3、5天 ,MCM促RN存活率分别为 40 7%、3 9 9%和 5 1 2 %。结论 巨噬细胞能分泌某些促进RN存活和生长的营养因子。
Objective To evaluate whether macrophage conditioned medium(MCM) can promote the retinal neurons(RN) survival and growth in vitro.Methods The retinae of postnatal 2~3 days Sprague Dawley rats were dissociated into cell suspension with trypsin digestion.Divided into the day 1,3 and 5 culture periods and each period farther divided into control group and MCM group. MTT colorimetric microassay was used to quantify the OD value of living cells in each well.Results The RN in the well coated by murine tail collagen tissue grew very well and some cells processed axons.MCM can promote significantly RN survival.At day 1,3 and 5,the RN survival rate promoted by MCM were 40.7%, 39.9% and 51.2%,respectively.Conclusion Macrophage can excrete some trophic factors promoting RN survival and growth.
