摘要
建立了 RP- HPLC法测定水飞蓟宾在人血浆中浓度的方法。用乙腈沉淀血浆蛋白 ,离心后 ,取上清液 10 μl进样分析。色谱条件 :C18柱 (10 μm) 2 2 0 mm× 4 .6 mm;流动相为 :V(甲醇 )∶ V(水 )∶ V(0 .5mol· L-1磷酸二氢钾 ) =10∶ 10∶ 1(用 0 .2 mol· L-1磷酸调 p H=4 .0 ) ;流速为 1.0 ml· min-1;检测波长为 2 90 nm。结果水飞蓟宾 A、B平均回收率分别为 98.96 %、97.93% ;日内 RSD分别为3.0 8%、2 .15% ;日间 RSD分别为 4 .4 8%、2 .6 4 %。
To find out a RP HPLC method for determining silybin in human plasma with acetonitrile as protein precipitation,the concentrations of silybin were assayed on a C 18 column and detected at 290nm with a mobile phase consisting of methanol water potassium dihydrogen phosphate solution (10∶10∶1) adjusted to pH=4.0 by adding phosphoric acid .The results showed that the average recoveries were 98.96% for silybin A and 97.93% for silybin B.The intraday precisions were 3 08% for silybin A and 2.15% for silybin B.The interday precisions were 4.48% for silybin A and 2.64% for silybin B.
