摘要
目的:探讨四妙勇安汤对动脉粥样硬化(AS)斑块内血管新生的影响及作用机理。方法:随机将40只日本大耳白兔分为对照组10只,实验组30只。对照组给予普通饲料,实验组利用复合因素制备AS易损斑块模型,8周时实验组随机分为模型组、辛伐他汀组、四妙勇安汤组,第24周取材。主动脉切片进行病理形态学观察,测量内膜厚度(IT)、纤维帽厚度(FCT)、纤维帽厚度与内中膜厚度比(FCT/IMT)、脂核与斑块面积比(LCA/PA);免疫组化方法检测CD31、血管内皮生长因子(VEGF)、P38 MAPK水平;RT-PCR方法检测VEGF mRNA表达。结果:与模型组比较,四妙勇安组IT降低,FCT、FCT/IMT增加;CD31、VEGF、P38MAPK水平及VEGF mRNA表达显著下降。结论:四妙勇安汤可稳定AS斑块,抑制斑块内血管新生,其机制可能与抑制斑块内VEGF和P38MAPK表达有关。
Objective: To observe the effect of Simiaoyongan Decoction(SYD) on angiogenesis in rabbit's Atherosclerosis(AS) plaque.Methods: 40 Japanese white rabbits were randomly divided in two groups,that is,the control group(n=10,normal diet) and the experiment group(n=30),in which AS vulnerable plaque model was established by feeding high-fat diet,injecting Bovine Serum Albumin(BSA) and balloon catheter overstretching on abdominal aorta artery.At the 8th week,30 animals in experiment group were randomly divided in three groups,that is,model group(n=10),SYD group(n=10),simvastatin group(n=10).The rabbits were sacrificed at the 24th week.Pathological changes were observed by HE,oil red O and Masson staining,and then measured intimae thickness(IT),fiber cat thickness(FCT),FCT/IMT,lipid core area to plaque area ratio(LCA/PA).CD31,VEGF,P38 MAPK protein were detected with immunohistochemistry(IHC),and VEGF mRNA signal was assessed by polymerase chain reaction(PCR).Results: IT in SYD group was lower and FCT,FCT/IMT levels were higher than that of the model group.Compared with the model group,the levels of CD31,VEGF,P38MAPK in the SYD group decreased,and VEGF mRNA expression was inhibited by SYD(P0.01or P0.05).Conclusion: SYD could suppress angiogenesis in AS plaque to stabilize AS vulnerable plaque.It may act by decreasing P38 MAPK,VEGF protein and VEGF mRNA expressions.
