摘要
采用渗透振扰破壁法、硫酸铵分级沉淀、乙醇分级沉淀、DEAE-纤维素层析等步骤提取和纯化重组L-门冬酰胺酶,并优化了提纯过程的条件,提取总回收率高于50%,产品纯度经高效毛细管电泳检测为单一峰,具有工业生产前景。
A combined technique of permeation vibration, (NH 4) 2SO 4 fractionation, ethanol fractionation and DEAE cellulose chromatography was used to extract and purify the recombinant L asparaginase Ⅱ.The total recovery of enzyme was greater than 50% by optimizing these procedures.HPCE analysis gave a final product of a single peak.
