摘要
目的采用高效液相色谱法研究并建立牦牛皮胶氨基酸类成分的指纹图谱,为牦牛皮胶的质量控制提供有效的方法。方法采用柱前衍生-RP高效液相色谱法,梯度洗脱进行分离,固定相为Agilent Analytical XDB-C18,(4.6 mm×250mm,5μm);流动相为pH6.4的醋酸钠缓冲液(A)-60%乙腈(B),检测波长为360 nm,体积流量为1.0 mL/min,分析时间为48 min。结果 10批样品具有较高的相似度。天冬氨酸、谷氨酸、丝氨酸、甘氨酸、苏氨酸、脯氨酸、丙氨酸、缬氨酸、甲硫氨酸、异亮氨酸、亮氨酸、苯丙氨酸和赖氨酸的平均质量分数分别为5.58%、10.49%、3.35%、23.25%、1.74%、14.19%、8.98%、2.48%、0.29%、1.6%、3.07%、2.12%和3.57%。结论该方法稳定可靠,可为其氨基酸质量的全面控制提供参考。
AIM To develop an HPLC method for controlling the quality of yak collagen according to the amino acids constituents.METHODS The amino acids constituents were analysed by Pre-column derivatization reversed phase high performance liquid chromatography.Chromatographic conditions were as follows: Agilent Analytical XDB-C18 column(4.6 mm×250 mm,5 μm),mobile phase was natrium aceticum buffer solution(A)(pH = 6.5)-60%acetonitrie(B),the detective wavelength was set at 360 nm,the flow rate was 1 mL /min and the analytical time was 48 minutes.RESULTS The result showed that the amino acids constituents had a high similarity.The contents of Asp,Glu,Ser,Gly,Thr,Arg,Ala,Val,Met,Ile,Leu,Phe and Lys were 5.58%,10.49%,3.35%,23.25%,1.74%,14.19%,8.98%,2.48%,0.29%,1.6%,3.07%,2.12% and 3.57%,respectively.CONCLUSION The method has good reproducibility and stability,and can be served as the means to control the quality of yak collagen.
