脓毒症大鼠血管内皮损伤因子的变化及辛伐他汀对血管内皮的保护作用研究 被引量:4

摘要 目的 探讨脓毒症大鼠血液中血管性血友病因子(vWF)、血小板活化因子(PAF)的变化规律及辛伐他汀对血管内皮的保护作用.方法 将54只SD大鼠随机分为脓毒症组(予腹腔注射0.9%氯化钠注射液,3h后予注射内毒素、制模后1、3、6、12h各6只)、辛伐他汀组(制模后1、3、6、12h各6只)和正常对照组,分别检测各组大鼠血vWF、PAF的水平.结果 脓毒症组大鼠在注射6h时的血vWF水平较正常对照组升高(P<0.05);辛伐他汀干预组6h时的血vWF水平较脓毒症组降低(P<0.05).脓毒症组在1、3、6、12h时的血PAF水平均较正常对照组明显升高(均P<0.01),辛伐他汀干预组上述时段的血PAF水平均较脓毒症组降低(均P<0.01),辛伐他汀组和正常对照组上述时段的血PAF水平的差异均无统计学意义(均P >0.05).结论 脓毒症大鼠血管内皮细胞受损大量释放vWF和PAF,使两者水平增高;辛伐他汀对脓毒症大鼠血管内皮具有保护作用,与其可降低血vWF、PAF水平有关. Objective To investigate the trend of change of vWF and PAF in blood in vibrio vulnificus septic rats,and detect the protection effects of simvastatin for endothelial cell. Methods To make sepsis model ,drug intervention model and normol model I in rats,detect the level of vWF and PAF in blood by ELISA. Results Compared with normal control group, vWF expression of sepsis mode increased markedly at 6h in sepsis model (P〈0.05),and compared with sepsis mode,vWF expression of intervention model reduced have statistical significance (P〈0.05);compared with normal control group, PAF expression of sepsis mode increased markedly at 1 h,3h,6h, 12h ( P〈0.01 ), compared with sepsis model, PAF expression of intervention model reduced markedly at 1h,3h,6h, 12h, compared with normal control group, PAF expression of intervention model increased have no statistical significance (P 〉0.05). Conclusion In sepsis, vessel endothelial cell was injured by excessive inflammatoryfactors, vWF, PAFwere released from endothelial cell. state.Simvastatin can protect endothelial cell,decrease greatly the level of vWF, PAF in blood.
