机械牵张力对人成骨样细胞糖链影响的初步研究

摘要 目的:研究机械牵张力对人成骨肉瘤MG-63细胞糖蛋白糖谱表达的影响。方法:以Flex-cell细胞应力培养系统对人成骨肉瘤MG-63细胞施以形变率为12%,加载时长分别为4、8、12、24 h的周期性张应变,利用凝集素芯片技术筛选出糖蛋白糖谱中差异表达的糖蛋白糖链,比较不同加载时长下成骨样细胞表面糖谱的变化。结果:8 h加力组表达差异最明显,在加力后实验组凝集素Jacalin、LEL、GSL-I、DBA、RCA、VVA、ACA的亲和作用增强,而对凝集素WFA、EEL、AAL、LCA、STL的亲和作用减弱,提示在机械牵张力作用下成骨样细胞表面可能出现增多的黏蛋白T/Tn抗原;I-抗原;N-乙酰半乳糖胺(包括α1-3链接方式),α及β链接半乳糖结构和减少的岩藻糖α1-2链接半乳糖;核心岩藻糖;N-乙酰葡糖胺结构等。结论:通过凝集素芯片技术研究发现受机械应力刺激后的成骨样细胞表面糖链变化与时间呈相关性,可推测在加载8 h后成骨样细胞已具备了某些骨化的早期表现。在加载4 h后成骨样细胞便出现对信号转导的重要调节修饰,为研究骨改建信号转导发生机制提供了糖组学理论支持。 AIM: To analyze the glycoprotein glycan profiles of MG63 osteoblastic cells under different mechanical strains.METHODS: MG-63 osteoblastic cells were loaded with mechanical strain loading by using Flexcell strain loading system.At 4h,8h,12h and 24h after loading,total protein was extracted from the loaded cells and control cells,labled with Cy3 fluorescent dye,then incubated with the lectin microarray.Glycoprotein glycan profiles were analyzed to screen differently expressed glycans.RESULTS: Mechanical strain stimulated MG-63 osteoblastic cells showed significant difference in lectin microarray as compared with non-stimulated cells.CONCLUSION: The lectin microarray results indicate that MG-63 osteoblastic cells exhibited time-dependent glycoprotein glycan profile changes under mechanical strain.Lectin microarray technique is an effective technique to investigate mechanisms of osteoblasts in bone remodeling.
