摘要
[目的]研究亚甲基蓝/光化学法对模拟受感染牛奶中大肠杆菌的灭活作用。[方法]以模拟感染了大肠杆菌的磷酸盐缓冲液(PBS)为参照系,通过平板菌落计数法观察不同浓度的亚甲基蓝(MB)和不同光照时间的溴钨灯光源对模拟感染大肠杆菌的不同浓度牛奶的光化学法灭菌效果。[结果]0.5×10-1μg/ml的亚甲基蓝在光功密度为200 mW/cm2的溴钨灯光照10 min情况下可以杀灭PBS中99%以上的大肠杆菌;而在同样光源下2,.0μg/ml的亚甲基蓝,光照10 min方可杀灭牛奶中99%以上的大肠杆菌。[结论]亚甲基蓝/光化学法对模拟受感染牛奶中的大肠杆菌具有良好的灭活作用,且溶液的光通透性可能是影响光化学法灭菌技术效果的一个重要因素。
[Objective] To study the methylene blue/photoinactivation against Escherichia coli in mock-infected milk.[Method] With the reference of mock-infected phosphate buffered salin(PBS),the effects of different concentrations of methylene blue(MB) and different illumination time of tungsten bromine lamp on the antibacterial effect of E.coli in milk were studied by plate culture count assay.[Result] 0.05 μg/ml Methylene blue illuminated by 200 mW/cm2 tungsten bromine lamp for 10 min would kill more than 99% E.coli cells in the PBS;but it needed 2 μg/ml MB illuminated for 10min to kill the same amount of E.coli in milk.[Conclusion] It was experimentally verified that Methylene blue-based photoinactivation was good in killing the E.coli in milk.Results also supported the idea that the light transmittance of solution was an important factor to determine the effect of photoinactivation.
