摘要
为定位检测日本乙型脑炎病毒(JEV)在试验小鼠脑内分布情况,本研究建立了JEV的小鼠感染模型,取发病死亡小鼠的脑组织制作石蜡切片,利用抗JEV单克隆抗体,采用免疫酶组化方法对抗原进行组织学定位检测。试验结果表明,所建立的间接免疫酶组化方法灵敏度高,特异性好,证明了JEV选择性感染神经元,抗原阳性表达主要集中在脑干、神经节及大脑皮质。
To localize and detect the distribution of Japanese encephalitis virus(JEV) in the brain of experimental mouse,we established JEV infection model in mouse and produced paraffin sections using brain tissue.We established indirect immunohistochemical method with monoclonal antibodies of JEV to localize and detect the antigen.Experiment results showed that the method had a high sensitivity and specificity,it demonstrated that JEV infect neurons selectively,the antigen expression were mainly in the brainstem,ganglia and cortex.
