黄颡鱼仔稚鱼发育过程中的胰蛋白酶原基因表达 被引量:3

摘要 采用RT-PCR和实时荧光定量PCR方法,得到黄颡鱼(Pelteobagrusf ulvidraco)消化系统胰蛋白酶原基因cDNA,并且对其早期发育过程中仔稚鱼(1~3日龄)消化系统胰蛋白酶原基因表达进行检测和定量。结果表明,黄颡鱼消化系统中胰蛋白酶原的基因表达起始于1日龄,1~6日龄表达量快速增加、6~23日龄表达量呈起伏式增加、23~30日龄表达量显著增加。该结果同时表明使用实时荧光定量PCR的方法检测黄颡鱼胰蛋白酶原基因的表达量,能反映其肠道消化功能在发育过程中的变化。建议黄颡鱼仔稚鱼转饵时间在23日龄后。 The first-strand cDNA sequence encoding pancreatic trypsinogen (TRY) gene was isolated from yellow catfish (Pelteobagrusfulvidraco) larvae (1-30 dph, day post-hatching) by RT-PCR, and using real-time PCR to detected and quantitated the trypsinogen gene expression. The results showed that the onset of trypsinogen gene expression in P. fulvidraco larvae was 1 dph and the trypsinogen gene expression increased significantly from 1 to 6 dph,increased fluctuantly from 6 to 23 dph and rose sharply from 23 to 30 dph. The experimental results demonstrated that the detection and quantitation of trypsinogen gene expression in P. fulvidraco larvae would accurately reflect the process that digestive functioning developed in intestinal tract. And we suggested that weaning time of P. fulvidraco larvae would be much better after 23 dph.
