从黄花菜(Hemerocallis minor Mill.)花粉粒中分离出生殖细胞,在含多种附加物以及花药作饲养物的 K,或 MS 琼脂糖培养基中进行离体培养,部分细胞发生一次核分裂,形成二核细胞;其中少数诱导了二次核分裂,形成3—4核细胞,个别细胞中似乎同时发生胞质分裂形成二细胞结构。诱导频率平均为3.27%,最高达11.46%。显微观察表明,生殖细胞呈纺锤、椭圆和圆球等形状,它们均能启动核分裂,但以圆球形细胞效果最佳。第一次核分裂有均等与非均等分裂二种方式,二者出现的机率相近。这是分离的生殖细胞在培养条件下启动发育的首次报道。
The generative cells isolated from pollen grains of Hemerocallis minor Mill.were cultured in modified K_3 and MS agarose media surrounded by liquid media containing anthers as a feeder.Microscopical observations on a total of 5961 cells revealed that nuclear divisiohs oc- cured leading to the formation of 2-4 nuclei within one cell.The average induction fre- quency among 14 culture experiments was 3.27% and the 11.46% as the highest.The generative cells varied considerably from spindle,oval to spherical shape.Among them the spherical cells appeared easier to be triggered.There were equal and unequal first nuclear divisions with almost equal chance of their occurence.A few of the cells passed through the second nuclear division,producing 3-4 nucleate ceils.Cytokinesis resulting in 2-celled structures seemed to be induced in rare cases.This is the first experimental attempt to culture isolated generative cells and the results show the potential of free generative cells surviving and de- veloping under In Vitro conditions.
