肝素对大鼠肝切除术后肝再生的影响

摘要 目的观察普通肝素对大鼠50%肝切除术后肝再生的影响。方法健康雄性Wistar大鼠54只,体重为230~290g,随机分为:A、B、C组。A组为50%肝切除术后3h皮下给予生理盐水(4ml/kg),B组为50%肝切除术后3h皮下给予肝素(1000U/kg),C组为假手术组(术后给予同剂量生理盐水)。各组在分别在术后72、120、168h随即处死6只,秤取肝脏重量,计算肝再生指数,机械法制肝脏单细胞悬液,碘化吡啶PI染色后流式细胞仪上机,ModFit LT forMacV.3.0软件分析,计算肝细胞增殖指数。结果 A、B两组肝切除术后72h、120h肝再生指数比较有统计学差异(P<0.05),而两组间术后168h肝再生指数无统计学差异(P>0.05)。A、B两组肝切除术后72、120、168h肝细胞增殖指数计较有统计学差异(P<0.05)。A与C,B与C各时间段流式细胞仪检测细胞增殖指数差异明显(P<0.05)。结论普通肝素(1000U/kg)可促进大鼠50%肝切除术后肝脏再生。 To investigate the effects of heparin on hepatic regeneration after 50% hepatectomy in rats. Methods Fifty -four Wastar rats (230 -290g) were randomly divided into three groups (A, B, C). Fifty percent of hepatectomy was performed in selected rats in group A and B. Heparin(1000 U/kg) and saline were given after partial hepatectomy through the subcutaneouly at a 24h interval befor sacrifice in group A and B respectively. Sham operation was performed in group C. The same volume of saline were given after opera- tion through the subcutaneouly at a 24h interval in group C. Six rats were randomly sacrificed in every group. The wet liver weights were measured separately,and the hepatocyte suspension was made. Porliefrate index(PI) was detected by flow cytometry. Results There were differences in hepatic regenerative ratio between group A and B three and fivedays after operation (P 〈 0.05). There was no differences in hepatic regenerative ratio between group A and B three and five days after operation (P 〈 0.05). There were conspicuous differences in PI index between group A and C,between B and C (P 〈 0.05). Conclusion Heparin (1000U/kg) given through the subcutaneously can significantly accelerate hepatic regeneration after 50% partial hepatectomv in rats.
