The relation of hydroxylase and covalently wall-bound peroxidase with the cell expansion of cucumber cotyledons was investigated,
The excised and aerated cotyledons of cucumber grew in basal medium (1/15 mol/L Na^+-K^+ phosphate buffer conraining 10 ppm 6-BA, pH 6.0 ) for 36 h in the dark. The covalently well-bound peroxidase activity increased with the increase of cell expansion of cucumber cotyledons (Fig. 1).
α, α'-dipyridyl is an inhibitor of hydroxylase. At the concentrations used, i. e., 0.1, 1 and 10 mmol/L, it inhibited both the activity of covalently wall-bound peroxidase and cell expansion of cucumber cotyledons (Table 1).
It was shown that when ascorbate was used as an inhibitor of peroxidase, the cell expansion of cucumber cotyledons was inhibited along with the inhibition of peroxidase activity (Table 2).
Therefore, the covalenfly wall-bound peroxidase possesses the function of peptidylproline-hydroxylase, and proline hydroxylation is essential to cell expansion.
The results of treatment with exogenous IAA showed an interaction between peroxidase and IAA.
