摘要
目的开发一种血吸虫病快速免疫诊断方法。方法用国产胶体染料 Ｄ－１标记日本血吸虫可溶性虫卵抗原（ＳＥＡ），以此标记物与血吸虫病人血清中的抗血吸虫抗体反应，再用点有羊抗人ＩｇＧ（第二抗体）的硝酸纤维层析膜以层析法捕获标记抗原与抗体的复合物。结果用试纸条法检测急性血吸虫病人血清３０份、慢性血吸虫病人血清８４份，其敏感性分别为９６．７％和９４．０％，检测健康人血清６０份，其特异性为９６．７％，检测肝吸虫病人血清和肺吸虫病人血清各２０份，其交叉反应率分别为 １０ ０％和 ７０． ０％．检测结果与常规ＥＬＩＳＡ法相似。标记染料在室温下至少可保存 ３个月活性不变。结论胶体染料试纸条法操作简便、快速，不需特殊仪器，适用于现场大规模检测。
Objective To develop a rapid, simple immunodiagnosis assay for schistosomiasis. Methods Soluble egg antigen of Schistosoma japonicum (SEA ) was conjugated with a blue colloidal dye. D-1 produced in China. The conjugated antigen could react with the antibodies in the patients' sera of schistosomiasis. 'The antigen-antibodies complex could be captured by anti-human IgG ab sorbed on the strip of nitrocellulose membrane by means of immunochromatography . Results The sensitivity of the colloidal dye strip immunoassay(:DSIA ) was 96' 7 % in 30 cases 'of acute schistosomiasis and 94. 0% in 84 cases of chronic schistosomiasis. Tile specificity of foe assay was 96. 7 % In 60 healthy objects. Tile cross reaction were ic% In 20 cases of clonorchiasis and 70. 0% in 20 cases of paragonimiasis. The results were similar ic that detected by routine ELISA. The ant lgen dye-labelled was stable at room temperature for at least 3 months. Conclusion 'The colloidal dye strip immunoassay provided high sensitivity and good specificity for detection of schistosomiasis and the assay was rapid, simple and did not need any equipment, it was inure useful for large scale de tection of schistosomiasis in endemic fields compared with other immunoassays.
